Abstract:

Objectives- To isolate, culture-expand and differentiate mesenchymal stem cells from ovine bone marrow and determine their culture requirements for high expansion rate. Design- Experimental study. Animals- Five Shal sheep. Procedures- In this study, ovine marrow cells were plated and culture expanded through 3 successive subcultures. The resultant cells were then plated at differentiating conditions into bone, cartilage and adipose cell lineages in order to verify their MSC nature. Furthermore, we determined the culture requirements of the cells in terms of FBS (fetal bovine serum) concentration and initiating cell seeding density. In this study, also population doubling time (PDT) was determined for the isolated cells. Results- According to our observations, MSCs from ovine marrow appeared to be fibroblastic in appearance. They were easily able to differentiate into bone, cartilage and adipose cell lineages as it was evident in RT-PCR analysis of specific gene expression and specific staining of differentiated cells. According to our findings, the cells indicated extensive proliferation when being cultivated at 100 cells/cm2 in a medium with 15% FBS. Ovine MSCs possessed a relatively short population doubling time (24.94±2.67 hours). Conclusions and Clinical Relevance- Fibroblastic cells from ovine bone marrow are able to undergo extensive proliferation and capable of differencing into three skeletal lineages, hence they are MSCs that are appropriate for cell therapy experimentation

Harvesting of bone marrow mesenchymal stem cells from live rats and the in vitro differentiation of bone marrow mesenchymal stem cells into neuron-like cells

In the bone marrow, there are certain populations of stem cell sources with the capacity to differentiate into several different types of cells. Ideally, cell transplants would be readily obtainable, easy to expand and bank, and capable of surviving for sufficient periods of time. Bone marrow mesenchymal stem cells (BM-MSCs) possess all of these characteristics. One of the most important benefi...

Differentiation Potential of Nestin (+) and Nestin (-) Cells Derived from Human Bone Marrow Mesenchymal Stem Cells into Functional Insulin Producing Cells

The feasibility of isolating and manipulating mesenchymal stem cells (MSCs) from human patients provides hope for curing numerous disease and disorders. Recent phenotypic analysis showed heterogeneity of MSCs. A nestin progenitor cell is a subpopulation within MSCs which plays a role in pancreas regeneration during embryogenesis. This study aimed to separate nestin (+) cells from human bone mar...

Osteogenic Differentiation of Rat Mesenchymal Stem Cells from Adipose Tissue in Comparison with Bone Marrow Mesenchymal Stem Cells: Melatonin As a Differentiation Factor

Background: Adipose-derived stem cells (ADSC) could be an appealing alternative to bone marrow stem cells (BMSC) for engineering cell-based osteoinductive grafts. Meanwhile, prior studies have demonstrated that melatonin can stimulate osteogenic differentiation. Therefore, we assayed and compared the melatonin effect on osteogenic differentiation of BMSC with that of ADSC. Methods: Mesenchymal...

Ex vivo Expansion and Differentiation of Mesenchymal Stem Cells from Goat Bone Marrow

Objective(s) Mesenchymal stem cells (MSCs) from large animals as goat which is genetically more closely related to human have rarely been gained attentions. The present study tried to isolate and characterize MSCs from goat bone marrow. Materials and Methods Fibroblastic cells appeared in goat marrow cell culture were expanded through several subcultures. Passaged-3 cells were then different...

comparison of proliferative and multilineage differentiation potential of sheep mesenchymal stem cells derived from bone marrow, liver, and adipose tissue

background: despite major progress in our general knowledge related to the application of adult stem cells, finding alternative sources for bone marrow mesenchymal stem cells (mscs) has remained to be challenged. in this study successful isolation, multilineage differentiation, and proliferation potentials of sheep mscs derived from bone marrow, adipose tissue, and liver were widely investigate...

comparison of differentiation potential of male mouse adipose tissue and bone marrow derived-mesenchymal stem cells into germ cells

background: recent publications about differentiation of stem cells to germ cells have motivated researchers to make new approaches to infertility. in vitro production of germ cells improves understanding differentiation process of male and female germ cells. due to the problem of using embryonic stem cells (esc), it’s necessary the mentioned cells be replaced with some adult multi-potent stem ...
